Temperature-Controlled Expression of a Recombinant Human-like Collagen I Peptide in Escherichia coli

Collagen is the functional protein of skin, tendons, ligaments, cartilage, bone, and connective tissue. Due to its extraordinary properties, collagen has a wide range applications in biomedicine, tissue engineering, food, cosmetics. In this study, we designed fragment human type I (rhLCOL-I) expressed it Escherichia coli (E. coli) BL21(DE3) PlysS containing thermal-induced plasmid, pBV-rhLCOL-I. The results indicated that optimal expression level rhLCOL-I reached 36.3% total at 42 °C, soluble form. 7 L fermentation, yield purified was 1.88 g/L. Interestingly, pBV220-rhLCOL-I, excellently stable during fermentation process, even absence antibiotics. Functional analyses had capacity promote skin cell migration adhesion vitro vivo. Taken together, developed high-level low-cost approach produce fragments suitable for medical E. coli.
